Iago est déterminé à détruire Othello, un général vénitien réussi qui a promu un autre homme nommé Cassio au lieutenant au lieu de lui. Roderigo est amoureux d'une femme nommée Desdemona, qui a déjà épousé secrètement Othello. Iago accepte d'aider Roderigo à sortir d'Othello. Iago envoie Roderigo au père de Desdemona, Brabantio, qui accuse Othello de faire passer Desdemona avec lui par la sorcellerie. Desdemona défend Othello, et leur mariage est validé devant le duc. Othello, Iago, Desdemona, Cassio, Roderigo et la femme Emilia d'Iago épousent à Chypre pour combattre les forces turques qui ont atterri là-bas. Une fois que la fête arrive, ils découvrent que la flotte turque a été détruite lors d'une tempête. Iago reçoit Cassio en état d'ébriété et le provoque dans un combat avec Roderigo. Othello blâme Cassio pour le combat et le dépouille de son rang. Iago envoie alors Cassio pour demander à Desdemona s'il est possible de convaincre son mari de le réintégrer. Emilia, l'assistant de Desdemona lors du voyage, prend un mouchoir que Desdemona tombe. Iago le prend, sachant qu'il sera utile plus tard. Il convainc Othello que quelque chose se passe entre Cassio et Desdemona, et se dépeint comme la seule confidente fidèle d'Othello. Othello favorise Iago au lieutenant.Iago plante le mouchoir dans la chambre de Cassio et dit à Othello d'écouter pendant qu'il a eu Cassio pour parler de l'affaire. En réalité, il demande à Cassio son affaire avec une femme nommée Bianca, mais Othello manque le nom et pense qu'il parle de Desdemona. Il décide de tuer Desdemona et demande à Iago de tuer Cassio. Iago convainc Roderigo pour le faire pour lui, et Othello procède terriblement à Desdemona. Roderigo attaque Cassio alors qu'il part de Bianca, mais il est blessé; Iago (déguisé) attaque Cassio et prétend ensuite arriver sur scène pour aider. Iago tue Roderigo pour le garder silencieux, et accuse Bianca de s'arranger pour tuer Cassio...
