About the Book

Translationally Controlled Tumor Protein (TCTP) ist ein wachstumsassoziiertes Protein, das in einer Vielzahl von Organismen von der Hefe bis zum Säugetier ubiquitär vorhanden ist. Es ist sogar eines der 20 am häufigsten in der Zelle exprimierten Proteine. TCTP wurde ursprünglich in einer Ehrlich-Ascites-Tumor-Zelllinie identifiziert, daher der Name. Später wurde nachgewiesen, dass TCTP auch in fast allen normalen Zellen vorhanden ist. TCTP ist auch unter den Bezeichnungen IgE-abhängiger Histamin-Releasing-Faktor (HRF), p23/p21 und Fortilin bekannt. TCTP hat ein Gewicht von etwa 20-25 kDa. Die erste Struktur von TCTP wurde 2001 mittels NMR-Spektroskopie aus Schizosaccharomyces pombe gelöst. Die Vorhersage der Tertiärstruktur von Proteinen wird zur Entwicklung von Modellen der Proteinstruktur verwendet, wenn die Beschränkungen durch Röntgenbeugung oder NMR-Spektroskopie nicht verfügbar sind. Bei der Tertiärstrukturvorhersage von Proteinen handelt es sich um einen bioinformatischen Ansatz, bei dem versucht wird, auf der Grundlage des Vorwissens über die Proteinstruktur eine neue Struktur zu erzeugen. Zur Vorhersage oder Modellierung der 3D-Struktur von Proteinen werden drei verschiedene Methoden verwendet: Homologiemodellierung (oder vergleichende Modellierung), Threading und Ab-Initio-Methode. Die 3D-Struktur von Proteinen ist für das Verständnis der Proteinfunktion notwendig.
