Xylose/Glucose-Isomerase ist ein wichtiges industrielles Enzym, das seit Jahrzehnten für die Umwandlung von Glucose in Fructose verwendet wird. Diese Forschung konzentriert sich auf die Produktion von Glukose-Isomerase aus Escherichia coli DH5α. Studien zur Optimierung der Fermentation zeigten, dass eine höhere Menge an Hefeextrakt und Trypton das Zellwachstum steigerte und die optimale Temperatur für das Wachstum 37°C betrug. Die teilweise Reinigung der Xylose-Isomerase erfolgte durch Wärmebehandlung und Ammoniumsulfat-Fällung. Bei 80 % des Salz-Cut-off-Wertes wurde die maximale Aktivität des Enzymkonzentrats erreicht. Die Xylose-Isomerase zeigte maximale Aktivitäten bei 75 °C und pH 8. Die Enzymaktivität bei optimalen Bedingungen betrug 5,0 U/min/ml für E. coli DH5α. Die Zugabe von Kobaltionen und Xylose erhöhte die Aktivität um das 10-20fache. Die spezifische Aktivität für E. coli DH5α betrug 60,5 nmol/min/mg Protein. Bei Verwendung von 100 mM D-Glucose als Substrat wurden nach 14 Tagen Inkubation nur 8 % des maximalen D-Fructosesirups produziert. Damit ist E. coli DH5α kein besserer Kandidat für die Produktion von High Fructose Syrup.
